Hematopoietic Stem Cells (HSCs) sit at the top of the hematopoietic hierarchy and are able to generate all the different blood cell lineages. This seminar will discuss on the embryonic site of origin of the HSCs. To better understand the cellular mechanisms of HSC development and specification, I generated a novel system to quantify the number of HSC precursors present at distinct stages of mammalian development. This novel approach was also employed to study the effect of aging in the clonal complexity of the hematopoietic system during unperturbed hematopoiesis. Its relevance in the acquisition of Clonal Hematopoiesis of Indeterminate Potential (CHIP) will be discussed.
