Rho GTPases are major regulators of the cytoskeleton and crucial for cell migration. For many years it was therefore believed that constitutive activation of Rho GTPase signaling will promote malignant progression of tumors. Cancer genome sequencing as well as genetic mouse models, however, could not confirm this and suggest a more complex role of Rho GTPase signaling in cancer.
